Interaction of Redox Probes and Ferrocene‐labelled Peptides with Lipid Bilayers Observed at Lipid Bilayer‐Modified Electrodes

چکیده

Glassy carbon electrodes (GCE) modified by a bilayer of phospholipids (BLM) have been used to monitor the passage redox probes and ferrocene-labelled peptides across lipid membranes an anionic (DOPG) or zwitterionic (globally neutral DPhPC) phospholipid. The gel-phase DPhPC showed strong association with like ferrocene methanol but expelled positively negatively charged species, as observed on cyclic voltammograms. Anionic DOPG underwent electrostatic interaction hexaamine ruthenium(III) complex, leading aggregates that caused swelling destructuration BLM. With ferrocene-tagged peptides, DOPG-modified GCE allowed observation voltammograms exhibiting both potential shift respect bare increased peak intensity, accounting for slow favored partition between BLM solution.

Interaction of Daptomycin with Lipid Bilayers: A Lipid Extracting Effect

Daptomycin is the first approved member of a new structural class of antibiotics, the cyclic lipopeptides. The peptide interacts with the lipid matrix of cell membranes, inducing permeability of the membrane to ions, but its molecular mechanism has been a puzzle. Unlike the ubiquitous membrane-acting host-defense antimicrobial peptides, daptomycin does not induce pores in the cell membranes. Th...

DNA interaction with freestanding cationic lipid bilayers

We study the interaction of ds-DNA molecules with freestanding and supported cationic lipid bilayers. DNA fragments of different lengths (5, 10, 20, and 48 kbp) were adsorbed on lipid membranes consiting mainly of zwitterionic DOPC with added fractions of cationic DOTAP (1...10 %). Freestanding bilayers were modeled using giant unilamellar vesicles of sizes >100 μm. Supported lipid bilayers wer...

Interaction of tea catechin (-)-epigallocatechin gallate with lipid bilayers.

A major component of green tea extracts, catechin (-)-Epigallocatechin gallate (EGCg), has been reported to be biologically active and interacting with membranes. A recent study reported drastic effects of EGCg on giant unilamellar vesicles (GUVs). In particular, EGCg above 30 microM caused GUVs to burst. Here we investigated the effect of EGCg on single GUVs at lower concentrations, believing ...

Differential interaction of Sophora isoflavonoids with lipid bilayers.

The mechanisms of some biological effects exerted by flavonoids (e.g. activity against lipid oxidation, multidrug resistance modulation) may involve their interactions with lipid bilayers. Due to variety of substituents attached to the flavonoid nucleus individual isoflavones significantly differ in their properties; in particular they may differently interact with membranes. For this reason we...
